Each individual model of Ride On Mowers is unique so it is important to check with the manufacturers operating instructions before operating any powered machinery, this is just a brief guide so that after you are familiar with your mowers operation it acts as a checklist of sorts to allow you to not miss anything that you may need to do in preparing your mower.

Remove all packaging that was supplied with the mower to ensure that all parts spin free and easy and that there are no areas that could get damaged by packaging being in the way or obstructing the operating radius of the mower. 
Ensure the mowers handlebars are in a correct operating position and that the handlebars are secured (Most mowers have turn-bolts that allow you to adjust the height, location and angle of the bars). 
Give the mower access to the power it requires.

Petrol Lawnmowers If the mower you own requires a petrol based power source then you will need to check if it requires any oil to be added to the fuel mix or if the oil is located in a different system, you need to fill the mower with enough fuel to successfully run the engine in and make sure that the engine has a sufficient oil/fuel ratio, typically when running a 2 stroke unleaded engine in you use a mix of around 20:1 (fuel:oil) and upon leaning the engine out after running in a mix of around 50:1. 
Electric Lawnmowers If your mower requires an electric power source connect the power distribution cable or PSU to an appropriate but secure power socket. 

Start the engine up and ensure it is running smoothly. 
Give the engine time to warm up (run the engine in by maintaining a low run speed if it is a new mower, or a mower that has been in storage a while). 
Mow your lawn!
